Development of Monoclonal Antibodies against Muc1/y Recombinant Protein Expressed in E.coli
نویسنده
چکیده
Cancer antigen 15-3 (CA 15-3) is a MUC1 peptide fragment widely used for the diagnosis of breast cancer. However, because of a lack of sensitivity and specificity, especially during early stages, CA 15-3 alone has not proved to be a reliable marker for early diagnosis of breast cancer. High preoperative concentrations of CA 15-3 are, however, associated with adverse patient outcome. Therefore, an emergent need still exists to find out secondary markers to enhance specificity of serum based detection of breast cancer. MUC1/Y a trans-membrane protein (Non-polymorphic), and a part of MUC1 gene devoid of tandem repeats array and its immediate flanking sequences has also been reported to be expressed in vivo by tumor cells and suggested to be a potential target both for epithelial tumor diagnosis and immunotherapy. In this study, we have expressed and purified recombinant human MUC1/Y from Escherichia coli BL21 (DE3) strain. We could develop a repertoire of monoclonal antibodies, which bound to our recombinant protein. However, most of the antibodies exhibited cross reactivity against normal human serum or His Tag. However, only 9A842G6 specifically reacted to recombinant MUC1/Y protein. 9A842G6 monoclonal antibodies exhibited fluorescence specifically with established breast cancer tissue sections and did not bind to fibroid adenoma sections. Specific reactivity of 9A842G6 mAbs towards MUC1/Y was also confirmed by immunoblotting. Therefore, our observations suggest possible use of MUC1/Y as a potential secondary marker for Breast cancer diagnosis along with CA153.

Production of Monoclonal Antibody against Prokaryotically Expressed G1 Protein of Bovine Ephemeral Fever Virus
Epitope-G1 of bovine ephemeral fever virus (BEFV) G glycoprotein has been genetically and antigenically conserved among various isolates of BEFV and only reacts with anti-BEFV neutralising antibodies. Therefore, it is a candidate antigen for development of the enzyme linked immunosorbent assay (ELISA) for serological identification bovine ephemeral fever (BEF)-infected animals. The aim of this ...
متن کاملProduction of monoclonal antibody against recombinant NS3 protein of bovine viral diarrhea virus (NADL strain)
Bovine Viral Diarrhea virus (BVDV) is an important viral pathogen of cattle causing several clinical syndromes. There are usually no pathognomonic clinical signs of BVDV infection. Diagnostic investigations therefore rely on serological detection and virus isolation. Nonstructural protein 3 (NS3) as immunogenic protein of BVDV is genetically and antigenically conserved among different isolates....
متن کاملImmunogenicity of chimeric MUC1-HER2 vaccine against breast cancer in mice
Objective(s): Breast cancer is one of the most common cancers in the world and is on the increase. MUC1 and HER2 as tumor-associated antigens (TAAs) are abnormally expressed to some extent in 75–80% of breast cancers. In our present research, a novel chimeric MUC1-HER2 (HM) protein was designed and used to study whether an immune response can be generated against these TAAs. In vitro analysis ...
متن کاملProduction of monoclonal antibodies against recombinant nucleoprotein of avian influenza virus, serotype H9N2
Background: Avian influenza viruses (AIVs) including the subtype H9N2 cause considerable financial losses to poultry industries. Rapid and accurate diagnosis of avian influenza (AI) infection is important in control and eradication programs. OBJECTIVES: The aim of this study was to produce monoclonal antibodies (MAbs) specific for the nucleocapsid protein )NP (of AIV H9N2 subtype to improve dia...
متن کاملFunctional Recombinant Extra Membrane Loop of Human CD20, an Alternative of the Full Length CD20 Antigen
Background: Targeting of CD20 antigen with monoclonal antibodies has become the mainstay in the treatment of non-Hodgkin's lymphomas and immunotherapeutic depletion of malignant B cells. Accessibility of antigen is one of the crucial factors in development of monoclonal antibodies against this antigen. One major problem in expression of full length CD20 is aggregation and misfolding. Therefore,...
